Clofarabine is a purine nucleoside analogue that is a lipophilic derivative of 2-Chloro-2'-deoxyadenosine (sc-202399). The advantage to using this compound compared to 2-Chloro-2'-deoxyadenosine is the increased stability of the acid. Studies suggest that Clofarabine contains the best qualities of Fludarabine (sc-204755) and 2-Chloro-2'-deoxyadenosine, with increased resistance to deamination and phosphorolysis. The compound functions by inhibiting intracellular ribonucleotide reductase and DNA polymerase, which suppresses DNA replication. Furthermore, Clofarabine displays greater affinity towards dCK (deoxycytidine kinase, dCyd), an enzyme essential in nucleoside phosphorylation.
